چکیده

THE study of bile pigment derivatives is important because these substances represent the end-products of haemoglobin metabolism. The destruction of haemoglobin both normal and pathological can thus be measured by quantitative determination of the urobilin and urobilinogen in urine and faeces. On the other hand, when pathological destruction of haemoglobin can be excluded, urobilinor urobilinogen-uria indicates failure of the liver to take up these substances from the portal circulation. Thus the test is valuable in the study of both blood and liver diseases. In the present work the Ehrlich-Neubauer colour reaction for urobilinogen with p-dimethylaminobenza]dehyde in HCI and the Jaffe-Schlesinger fluorescence reaction with zinc-acetate-alcohol for urobilin have been studied as qualitative urine tests. Neubauer [1903] identified urobilinogen as the reacting substance in Ehrlich's aldehyde test but described also positive reactions with other pyrrole derivatives such as reduced haematin and haemopyrrole. Thomas [1907] summarised the chemical properties of urobilin and urobilinogen and mentions indole and tryptophan as interfering substances. Fischer and Meyer-Betz [1911; 1912] first isolated urobilinogen from a pathological urine and assumed this substance to be identical with mesobilirubinogen prepared from bilirubin. Watson [1932; 1933] and Lemberg [1934] however stated that urobilinogen is not identical with mesobilirubinogen which differs from the former in its characteristic violet iron reaction. The quantitative estimation has been greatly improved by Charnass [1909] who first applied Ehrlich's test quantitatively instead of using the less stable urobilin reactions. Terwen [1924] devised a quantitative method introducing ferrous salts for the reduction of urobilin.

36. Coupled Oxidation of Ascorbic Acid and Haemoglobin 5. Studies on the Formation of Bile Pigments from Choleglobin and Verdohaemochromogen and on Their Isolation from Erythrocytes1

IN a preliminary paper [Lemberg, Legge & Lockwood, 1938] we have already reported the isQlation of biliverdin (dehydrobilirubin) and of biliviolin from. choleglobin solutions. Under remarkably mild conditions haemoglobin is thus transformed. into bile pigments; these conditions are such as are found in the cells of organs forming bile pigment, except that acid was used for the removal of the ir...

Heterogeneity of bile pigment conjugates as revealed by chromatography of their ethyl anthranilate azopigments.

1. Azopigments derived from conjugated bile pigments by coupling with the diazonium salt of ethyl anthranilate are analysed conveniently by quantitative t.l.c. or by column chromatography on CM-cellulose. 2. By chromatographic studies combined with a series of chemical tests six groups of azopigments were demonstrable in preparations from bile and from icteric urine of man. Azobilirubin and its...

Evaluating the Beneficial and Detrimental Effects of Bile Pigments in Early and Later Life

The heme degradation pathway has been conserved throughout phylogeny and allows for the removal of a pro-oxidant and the generation of unique molecules including bile pigments with important cellular functions. The impact of bile pigments on health and disease are reviewed, as is the special circumstance of neonatal hyperbilirubinemia. In addition, the importance of promoter polymorphisms in th...

Separation by thin-layer chromatography and structure elucidation of bilirubin conjugates isolated from dog bile.

1. A system for separation of bile pigments by t.l.c. and for their structure elucidation is presented. Separated bile pigments are characterized by t.l.c. of derived dipyrrolic azopigments. 2. At the tetrapyrrolic stage hydrolysis in strongly alkaline medium followed by t.l.c. demonstrates the presence of bilirubin-IIIalpha, -IXalpha and -XIIIalpha and allows assessment of their relative amoun...
